To be honest, the truth is no one really knows this apart from jagex them selves. I should think mouse tracking and all the milarki like that is quite abundant, unless the script it self is nearly a 100% repetitive then obviously bot tracking in that circumstance does become very viable to them. The only real way i should think they pick up on these things is the velocity of failed random events and such, also the most 100% way is by non player compliance with moderators, and filed player reports.
