So when you get manually picked out, at say experiments... Is there literally a JMod walking around and he talks to you or what? You get a chance to respond or are you just automatically toast?

Those are manual bans but there is system bans too. They track the path your walking, clicking, etc. The system bans you if you walk the same path, same tile all the time. 

Bot Busting = Manual.

Not sure if they just do it because you don't respond. It's been speculated they have behind the scene tools to get some additional information about your account.

 

Macroing (Minor/Major) are thought to be automatic.

I got a bot busting ban some bs.

i doubt they would talk to you, they just observe... it's pretty easy to tell even without the gear. i have been banned many times one of which included a main all stats back to 10 no msg in inbox =/
